The 23-year-old missed Saturday's win over Blackburn with the ankle problem and scans have confirmed the classy midfielder requires surgery on the injury.
The surgery is set to sideline Huddlestone for up to three months meaning he will not be back until February at the earliest.
A club statement read: "Following consultation with several independent ankle specialists, it has been determined that Tom Huddlestone will undergo surgery on his right ankle.
"The procedure is likely to rule the England international out of action for around three months."
The news is a major blow for Spurs boss Harry Redknapp as he now has to contend without his midfield kingpin for the rest of the Champions League group stage as well as a number of key Premier League games.
Huddlestone joins the likes of Jermain Defoe, Michael Dawson (both ligament damage), Jamie O'Hara (back) and Jonathan Woodgate (groin) as long-term casulaties this season for Spurs.
